IGF 1 LR3
IGF-1 LR3 (Insulin-like Growth Factor-1 Long Arg3) is a synthetic 83-amino acid analogue of human IGF-1, engineered with two structural modifications: substitution of glutamic acid at position 3 with arginine, and addition of a 13-amino acid N-terminal extension (Met-Lys-Arg extension sequence). These modifications dramatically reduce binding affinity for IGF binding proteins (IGFBPs 1–6) compared to native IGF-1, resulting in a prolonged half-life in serum-based assay systems due to reduced sequestration. IGF-1 LR3 signals through the IGF-1 receptor (IGF-1R), a receptor tyrosine kinase, activating downstream PI3K/Akt and Ras/MAPK signaling cascades in cell-based models. It has been widely used as a research reagent in studies of myoblast and satellite cell proliferation, protein synthesis pathway characterization, and insulin receptor substrate (IRS-1/IRS-2) phosphorylation kinetics in skeletal muscle and adipocyte cell culture systems. Researchers also employ IGF-1 LR3 in receptor binding competition assays and IGFBP displacement studies due to its reduced protein-binding profile relative to recombinant human IGF-1.
